摘要

In this study, we have reported a novel derivatization method for the specific and efficient derivatization of histones with NHS-propionate. Our method can be conveniently executed and meanwhile achieve excellent derivatization specificity and efficiency in a reproducible manner. A competitive inhibiting strategy was designed and then successfully used in our method to achieve the excellent specificity for derivatization. As our method can achieve above 90% conversion rate of histone peptides, it should be of great value in the relative and absolute quantification of histone modified peptides.
