ABC294640 was detected in tissues with and without CASI, also with CID + CASI to confirm fragmentation patterns in tissue and presence of drug. Drug distribution and metabolite analysis of ABC 294640 in orthotopic pancreatic tumor and other tissues is ongoing. SK2 inhibitor ABC294640 treatment in an orthotopic pancreatic mouse tumor model affected the levels of different phospholipid, ceramide and glycan species in the drug treated mice as determined by a full profile MS scan of these biomolecules tissue. Metabolic labeling with azido-sugars in Panc-1 cells combined with Click-it chemistry and MS identified potential glycoproteins modulated by ABC294640 treatment.
