1. MALDI-MS, LC-MS/MS and FIA-MS/MS can be effectively used to quantify gemini surfactants within PAM212 tissue culture matrix 2. Completed method validations demonstrate that MALDI-MS is superior to both LC-MS/MS and FIA-MS/MS MALDI-MS was superior for its linearity, achieving nearly 1000x linear range 3. MALDI-MS achieved the highest ULOQ and nearly achieved the optimal LLOQ of FIA-MS/MS 4. A drawback of the observed fragmentation of the gemini surfactant is that during quantification, MALDI-MS analysis would not be able to distinguish between in vivo/vitro analyte breakdown at that specific bond (Fig. 3) or fragmentation which occurs during ionization within the mass spectrometer
