1. We have confirmed our ability to sample in-situ from the membrane surfaces of marine invertebrates to obtain unique signatures for FA's; 2. We have verified our anion "sniffer" enrichment stage for quantitative recovery of FA's and diglycerides using spiked samples; 3. We have confirmed our ability to sample from the tentacle of a sea anemone to obtain representative membrane lipids and other yet unidentified components many of which contain nitrogen; 4. We observe 100's of unique components from these organisms, many of which are at low levels and difficult to interpret without increased sensitivity and specificity; 5. We are advancing the present work to measure the modulation of these FA and lipid signatures with external biotic and abiotic stresses; use of APCI to measure oxylipins will be included; 6. Further platform development will be necessary to achieve our objectives: (1) An accurate mass instrument with MS/MS will be required to improve interpretation; (2) Modifications are being made to the ion source to improve ion transmission; (3) Additional sample isolation channels for cations and zwitterions are being designed to meet our ultimate goal of a broad-based method for in situ metabolite and biomolecule characterization.
