1. A new untargeted lipid profiling methodology has been developed for studying C. elegansmutants involved in embryonic elongation (hypo- and hyper-contraction) with high resolution ESI-QqTOF-MS in both positive and negative ion mode 2. Data processing was conducted using multiple softwares included Markerview, LipidView, PeakView and MultiQuant. 3. Atotal of thirty lipid species were tentatively identified using LipidView software based on high resolution mass data and MS/MS fragments were shown to be statistically relevant as potential lipid markers of elongation perturbations inC. elegans. 4. Subsequent work will involve the validation of these lipid markers with more elongation mutants using a targeted strategy and direct nanospray infusion.
