By using fast scan speed MS spectrometers with high mass resolution, 331 proteins (1% FDR) were successfully identified in a single normal and cancerous samples. This result is currently the most number of proteins ever recorded in a 1D LC-MS/MS analysis of non-depleted plasma. Target biomarker peptides were successfully detected and quantified from both normal and patient plasma samples in a profiling workflow, where qual and quant analysis could be performed at the same time. Target peptides were additionally validated in MRM workflow showing better sensitivity.
