摘要

1.We found the key peptides of Fab-CR6261 located in 4 different loops reaponsible form the SC 1918/H1 binding. These peptides exhibited significantly different deuterium uptakes when the Fab was analyzed in complex with HA. 2. The paratope mapping results obtained by both HDX and X-ray crystallography agreed will on the binding regions at peptide level. 3. By comparing deuteration levels in overlapping peptides, we were able to narrow down the number of possible cintact sites. 4. Waters HDX technology using UPLC system and automated software is a rapiid alternative tool to probe for proteinprotein interaction studies.
