High-throughput Pipeline for the Purification and Identification of Desulfovibrio vulgaris Membrane Protein Complexes

The ability of the Gram-negative sulfate-reducing bacterium Desulfovibrio vulgaris (strain Hildenborough) to reduce heavy metals makes it an ideal candidate for bioremediation efforts. Knowledge of this organism's molecular level responses to environmental changes will be essential for accurately modeling its stress response pathways and developing strategies to optimize the application of D. vulgaris in the field. To obtain this knowledge we have developed a high-throughput pipeline for the isolation and identification of untagged membrane protein complexes. This "tagless" strategy will provide a global view of changes in membrane protein complexes from bacterial cultures grown under a variety of conditions. An additional advantage of the "tagless" approach is that it does not require genetic manipulation which may introduce artifacts.
