Detection of 3 amino acid substitutions (G102A, C1210A and G110A) in recombinant histone H3 and its mono-, di & tri-methylated isoforms by ESI-FTMS, combined with CAD, ECD, top-down proteome DB search and de novo peptide tag sequencing. Structural identification of the chemical analog of methyl Lys-9 -- aminoethyl cysteine through accurate mass measurement and generation of rational elemental composition, without prior information about the chemical modification.
