Enrichment and analysis of bacterial glycopeptides from total proteome extracts of Francisella tularensis

Preliminary gel based proteomics showed multiple protein spots to react with a commercial glycostain reagent. IP-NPLC method allowed enrichment of bacterial glycopeptides from total tryptic proteome digests. Subsequent MS analyses identified a bacterial glycopeptide, harbouring a 1156 Da modification. A combination of electron transfer dissociation and beta elimination showed this modification to be in O-linkage.
