SDS-PAGE-MALDI-TDS: Mass Spectrometric Top-Down Sequencing of Proteins Extracted from Polyacrylamide Gels

TDS allows fast and reliable characterization of both N- and C-termini of pure proteins. Mixtures of proteins as in reduced mABs require separation prior to TDS analysis. SDS-PAGE combined with protein purification allow TDS of separated standard proteins. High quality TDS spectra of light and heavy chains of mABs were achieved using this method and N-and C-terminal modifications were determined.
