Creation of a cocktail of 45 SIS peptides with relative abundances balanced to match their endogenous plasma concentrations. 45-plexed MRM assays with linear responses (R>0.999) over a 10~(4) concentration range and up to attomole-level detection sensitivity. Establishment of an automated robot workflow for sample preparation. First application of MRM standards cocktail to the quantitation of 45 proteins in a 40-patient CVD cohort., with 88percent of protein concentration measurements in all samples achieving CVs of analysis below 20percent. Using a multivariate classifier, most samples are correctly grouped. Further method development will extend this strategy to quantify additional 50 proteins related to CVD.
