A fully integrated autosampler/SPE-MS/MS system was successfully implemented into the PAMPA workflow. A generic solid phase extraction method was established that was successfully used with a large set of compounds from a wide chemical space. A custom workflow process was developed to complement the speed of SPE-MS/MS analysis. PAMPA permeability correlation plots generated for the comparison of LC/MS/MS to SPE-MS/MS yielded a slope of approximately 1 and a correlation coefficient of approximately 0.9 for 108 proprietary and literature compounds. The permeability results from traditional LC/MS/MS and SPE-MS/MS were deemed equivalent. The PAMPA attrition percentage was similar between LC/MS/MS and SPE-MS/MS.
