Four standard proteins, BSA, myoglobin, cytochrome c and ovalbumin, were first used for optimizing the microwave energy, temperature, incubation time. Compared to the commonly used protocols, it only takes 2 minutes for tryptic digestion while the sequence coverage for each protein was increased by approximately 20percent. When this optimized condition was applied to digest the proteins in gel bands, higher sequence coverage in a short time digestion without any other treatment were. Our results demonstrate the potential for the use of this method for the analysis of real time samples.
