In situ IRRAS has been developed as an effective technique for in situ real-time characterization of surfaces modified by SL/RL. For SL of [GS+2H]~(2+) onto FSAM, it shows linear accumulation of GS during deposition and stabilized signal after deposition. For RL of diamine ion onto NHS-SAM and COF-SAM, bond making/bond breaking processes were directly observed by recording the characteristic IRRAS bands; Relative intensity of the IRRAS band is determined by the orientation of the corresponding bond relative to the surface. Conformationally-selected peptide arrays could be prepared by SL of peptide ions onto SAM surfaces; the evolution of different conformations during SL was studied for the first time.
