The permethylation and stable-isotope labeling MALDI-TOF approach for the comparative N-glycome profiling of Drosophila is presented. Quantification capability of the method showed very good linearity (R~(2)>0.99) over a approx10 fold change in d_(0)- to d_(3)-permethylated glycan ratio in the m/z 1000-3000 range. High mannose N-linked oligosaccharides were observed to be the major type of N-glycans in both Drosophila bodies. The expression of specific N-glycans changed in relative abundance in Drosophila bodies as a function of age. The changes in N-glycome of Drosophila heads will be examined. The isomers of Drosophila N-glycome will be studied using IMS-MS in combination with molecular modeling.
