Proteomic Analysis of a Sinitang Uptake Response in the Radioresistant Bacterium Deinococcus radiodurans

In this study, it was found that a sinitang-treated culture exhibited a growth curve characterized by a very rapid exponential-stationary transition, while the untreated cells reached the early stationary phase synchronously (fig. 1). These results suggest the presence of a sinitang-sensitive mechanism for controlling cell division. To directly assess the effects of sinitang treatment on the global protein profiles of D. radiodurans, the differential proteomes among the control, sinitang-treated and Mn(II)-added cells were characterized at early stationary phase of D. radiodurans growth. Five proteins had been identified by MALDI-TOF-MS using peptide fingerprinting. Among them, three proteins (spot no. 2,4,7) were up-regulated by adding Mn(II) and sinitang, two proteins(spot no. 10 and 11) were down-regulated by Mn(II)-additive. Phosphopyruvate hydratase, one of the five identified proteins, was associated with glycolysis. These findings may offer valuable insights into the metabolic pathways affected by sinitang treatment in D. radiodurans cells and study the chemical constituents of sinitang.
