The function of alpha-synuclein and its role in Parkinson's disease (PD) has been heavily studied since mutations were discovered in several Greek and Sicilian families with a familial form of the disorder (1). Subsequently alpha-synuclein was determined to be a major component of Lewy bodies, the proteinacious inclusion bodies which are a hallmark of the sporadic disease (2). Lewy bodies contain a high concentration of nitrated tyrosine residues as determined by antibodies specific for 3-nitrotyrosine (3NT) (3). Utilizing the flexibility and sensitivity of MRM Mass Spectrometry we have quantified 3NT levels of alpha-synuclein and found a increase at a specific tyrosine using a PD model (MAO-B overexpression).
