Multiplexed quantitative proteomics analysis of mosquito midgut with Plasmodium oocysts and human blood background showed novel temporal changes in proteins. We have identified more than 200 Plasmodium falciparum proteins specifically from A. gambiae midgut after feeding parasite-infected blood meal The data from proteomic analysis using high accuracy mass spectrometry is valuable for genome annotations. This approach has been used to identify novel exons, splice variants and novel genes and these results are validated by comparative genomics and RT-PCR. Different mass spectrometry approaches such as CID, PQD, HCD for MS/MS level reporter ions quantitation needs to be thoroughly evaluated for accurate profiling of samples.
