Proteomic Quantification of the S-Nitrosylated Proteins in Parkinson Disease Paradigms

Parkinson's disease (PD) is a commonly occurred late-onset, progressive movement disorder, which is characterized by resting tremor, rigidity, and bradykinesia. Strong evidence suggests that nitrosative/oxidative stress results in the dysfunction of parkin and UCH-L1 via S-nitrosylation (a redox-based modification of protein specific cysteine thiol groups by signaling molecule nitric oxide (NO) forming S-nitrosylated-proteins, SNO-P) to regulate protein activity, and thereby contributing to the pathogenesis of the sporadic form of PD. Previously, these S-nitrosylated proteins were individually identified. We developed a gel-based proteomic strategy, named as NitroDIGE, to quantitatively investigate a subset of proteome of the S-nitrosylated proteins in PD.
