When two or more medications are administered concurrently, drug-drug interactions may take place that can affect their pharmacological response. These interactions may or may not result in altered therapeutic efficacy or increased toxicity. One of the main difficulties in understanding the practical relevance of drug-drug interactions in domestic animals is the dearth of in vivo clinical data. Most of the published information comes from in vitro experiments that provide very valuable mechanistic information and can be used to make reasonable predictions about possible or likely interactions in animals. In practice, however, many factors influence the clinical significance of drug-drug interactions; these include size and frequency of dose, duration of treatment, time-course of the interaction, drug therapeutic index, disease status, pharmacokinetic differences between interacting drugs, status of the immune system, inter-animal variability, etc.
