Over the years human apolipoprotein A-I (apo A-I), the major protein component of high density lipoproteins (HDL), has been intensely studied to understand the mechanism by which it protects atherosclerosis. Recent observations that administration of apo A-I_(Milano):phospholipid complexes causes regression in humans have given further stimulus to understanding the structure and function of apo A-I . Our approach over the years has been to study the nature of amphipathic helical motifs, the lipid associating domains present in this and other plasma apolipoproteins responsible for not only associating with phospholipids but also exerting antiatherogenic properties.
