MDR1 is a gene known as the multidrug resistance 1 gene. The new name for the MDR1 gene is ABCB1 (which stand for ATP-Binding Cassette subfamily 1 gene). This gene codes for a protein called P-glycoprotein (P-gp). P-gp was first identified three decades ago in chemotherapy-resistant cancer cell lines where it was found to pump a variety of chemotherapeutic drugs out of the cancer cell1. Because these chemotherapeutic drugs were no longer able to kill tumor cells that expressed P-gp, the tumor cells were described as multidrug resistant (MDR). The way that P-gp is able to cause resistance to many anticancer drags (rather than just a single class of drugs) is related to where it is expressed on the cell and how if functions.
