Column bioleaching of chalcopyrite was conducted at 33, 45 and 65□. After 220 days, the copper leaching rate at 65□ was 56.75%, whereas it was 38.50 and 51.35% at 30 and 45 □ respectively. Microbial community structure of both bacteria and archaea in the columns was analyzed using 16S rRNA gene clone library at day 220. Clone library results indicated that microbial community at the tested temperatures had both iron-oxidizers and sulphur-oxidizers, whereas iron-oxidizers were dominant at 33 and 45 □ and sulphur-oxidizers were dominant at 65 □. Leptospirillum ferriphilum were the dominant bacteria at 33 and 45 □. Uncultured sulfur-oxidizing symbiont bacteria were the dominant bacteria at 65 □. With regard to archaea community structure, Ferroplasma sp. were the only detected archaea at 33 □. Cultures similar to uncultured archaeon clone were the main archaea at 45 □. Metallosphaera sedula were the dominant archaea at 65 □. It suggested that different temperature determined different community structure.
