Traditional matrix assisted laser desorption ionization (MALDI) mass spectrometry (MS) can have spectral interferences in the low mass range due to matrix ionization and fragmentation. Whole cell analysis is especially difficult with MALDI-MS because of the requirement for matrix-sample cocrystalization and limitations on crystal size. Laser desorption ionization (LDI) on silicon nanopost arrays (NAPA) has been shown to yield meaningful metabolic information from biological cells and tissues. Here we show the analysis and imaging of cells and cell lysates of Saccharomyces cerevisiae and Bradyrhizobium japonicum using LDI-MS from NAPA.
