LC-MS analysis using PGC packed capillaries resulted in detection levels in the nanogram range, well over an order of magnitude less than in routine nucleoside analysis using microbore technology. Hyper retention of the methylated guanosines and their analogs is a concern that needs further investigation. The use of a modifier in the mobile phase may alleviate this retention and experiments are planned. The increased ionization efficiency and separation performance allowed the detection 2'-O-methyl adenosine, a previously undetected modification in the E. coli MRE 600 organism. Due to its mechanical robustness and relative chemical inertness, porous graphitic carbon has been shown to be an adequate substrate for capillary chromatography of RNA hydrolyzates.
