A new global LC-MS/MS method was developed to analyze FFA and oxidized FFA. 253 various oxidized FFA including tetracosanoids were identified in the contusional cortex after TBI in which 50 were confirmed by MS~2 analysis. Oxidized FFA levels increased early (1 hr) after TBI and decreased by 24 hr albeit remained higher than control. 49 oxidized DHA products were detected after TBI and the mediators such as 14-HDHA, 17-HDHA, 14-HpDHA, 17-HpDHA, protectin DX1, neuroprotectin, Resolvin D1 and D5 are significantly elevated up to 24 hr after the injury. The oxidized DHA-derived mediators are likely generated from infiltrating inflammatory cells as we have shown increased neutrophil recruitment into the pericontusonal cortex within the first 24hr after injury.
