Cancer progression is a complicated process and understanding the exact mechanism underlying these changes will provide invaluable knowledge in the diagnosis and therapeutic interventions against cancer. As the genomic read out, proteins play vital roles in various cellular processes and changes in protein composition in cancer cells may serve as important biomarkers and targets for cancer therapy. Here we utilized a mass spectrometry-based technique for the quantitative analysis of global proteome reprogramming between the primary human skin melanoma cell line, WM-115, and its matched metastatic cell line, WM-266-4, with stable isotope labeling by amino acids in cell culture (SILAC).We were able to elucidate the cellular mechanisms behind this skin melanoma progression via interrogating the differential protein expression level.
