Providing the benefit of both high resolu7on and high mass accuracy, Orbitrap-based mass spectrometry (MS) is a powerful tool for the characterization of intact proteins and their primary amino acid sequences. However, the adoption of Orbitrap-MS based technology is widely underu7lized due to the lack of low cost and easily implemented experimental techniques for most user facilities. Presented herein is a robust workflow for the characterization of intact proteins using high resolution MS and top-down MS on an Orbitrap Elite mass spectrometer.
