HNE modifications on specific amino acid residues is considered as a biomarker for many metabolic, hereditary, and age-related diseases. In the present study, we used lysozyme and BSA as model proteins to further investigate the formation of HNE-amino acid adducts. Using direct infusion to ESI-QTOF micro mass spectrometer, we were able to find HNE modifications on up to seven amino acid residues per molecule of lysozyme. Furthermore, using NanoAcquity UPLC coupled with QTOF Ultima API mass spectrometer, we also investigated the sites of these HNE modifications.
