The major focus of this study was on the development of a targeted proteomic method for assessing GTP-binding proteins at the proteome-wide scale. In particular, we established a spectral library with retention time and MS/MS information for peptides from over 135 GTP-binding proteins. In addition, we utilized an affinity-aided, mass spectrometry-based targeted proteomic workflow as an effective approach for globally profiling GTP-binding proteome. MRM-based analyses were conducted as a way to examine and fine-tune the effectiveness of the method in providing reliable targeted measurement.
