MALDI-TOF mass spectra of small tRNA fragments obtained from digestion of Transcript tRNAIle and Natural tRNAIle species by RNase T1 exhibit the expected ion peaks, confirming the nucleobase sequence. Unfortunately the position of some modification could not be identified. For SFC-MS, a screening of different columns was firstly done in order to determine the best stationary phases for the base peak separation of modified nucleosides. Solvent make-up and source ionization conditions were optimized, leading to the best sensitivity using the ESI source.
