Using isotopic labeling, MS3, and computational strategies, we were able to formulate a work flow that enabled the rapid identification of Amadori product modified peptides. In a test sample containing 12 spectra of Amadori product modified peptides, using only computational approaches, we were able to reduce 2929 total MS/MS spectra down to 36 spectra for manual interpretation. This combination of approaches allowed for the identification of 13 additional modifications on the peanut allergen, Ara h 1, that we had not been able to previously characterize.
