Long-term (> 24 h) dynamics of biomolecule concentrations are often of interest in cellular studies. As a result, separations-based monitoring of biomolecules would benefit from simple, robust, automated methods of sampling and detection of compounds. We have developed an automated method for sampling and injection of compounds for liquid chromatography-mass spectrometry (LC-MS) detection. The automated system can be coupled to cell chambers or reaction vessels to allow a diverse array of applications. The system has been used to quantitate intact proteins such as insulin, as well as small molecules such as drugs and metabolites. Additionally, the system has been applied to continuous reaction monitoring studies.
