Structural characterization of Lipid-A from Burkholderia pseudomallei and Burkholderia thailandensis was performed in this study using MALDI-MS and ESI-MS. MALDI-MS spectra were obtained in negative mode using 6-aza-2-thiothymine (ATT) as matrix. MS/MS experiments were performed to obtain more details about lipid A structure. Using MS and MS/MS data, we proposed structures of Lipid A species. Lipid A likely has a disaccharide backbone with two phosphate groups and two 4-amino-4-deoxyarabinoses attached to these phosphate groups. The disaccharide backbone is hepta-acylated with C_(12:1), C_(14:0) (2-OH), three C_(14:0) (3-OH), C_(16:0), and C_(18:0).
