Determination of Urea in Human Serum and Epithelial Lining Fluid Using LCMS; Benchmarking to a Traditional Diagnostic Colorimetric Kit

Serum can be stripped of endogenous urea utilizing a dialysis membrane. Mixture of HSA/saline serves as viable surrogate matrix for ELF Derivatization creates a larger, more non-polar molecule with added chromatographic retention and a selective transition. A lower limit of quantitation below the endogenous level can be achieved. A linear response over the range of 0.5 to 10 μg/mL in ELF and 5-100 μg/mL for dialyzed human serum can be achieved.
