The PhIP level in hair correlates to PhIP intake and the relationship is strengthened after adjusting for the melanin content. The reduction performed directly on the hair digest was impaired by the presence of the complex digest matrix. Comparing the three MS analyses methods, MS~3 mode employing HCD with a mass resolution of 120,000 yielded the cleanest mass spectra but lowest signal. Further instrument optimization is required to improve the signals.
