Direct elution of co-immunoprecipitated peptides is as reliable a tool for the identification of novel protein/peptide binding partners within the mTOR-Raptor signal transduction pathway. Our method should prove useful to study other pathways and proteins involved in T-cell activation such as TCR-zeta chain, SLP76, and CTLA-4. Neutral loss mass spectrometry method is a powerful tool for the identification of phosphorylated proteins/peptides. Signaling to mTOR appears to involve phosphorylation at multiple sites. Mutation studies will demonstrate the role these sites may have in T-cell activation and tolerance.
