Glycosylation site mapping using traditional approaches without isotopic labeling produces erroneous glycopeptide identifications; especially when performed on an instrument with lower mass accuracy. However, when isotopic labeling is incorporated and the labeled glycopeptides are analyzed using a mass spectrometer with accurate mass measurement capabilities, the frequency of random glycopeptide assignments is greatly reduced.
