Development of molecular biomarkers for lung cancer is needed for the assessment of reduced harm products. It has been previously observed that protein secretions increase during cancer metastasis (1,2). A global proteomic investigation of in vitro cell cultures representing lung cancer disease progression (BEAS-2B (nontumorigenic), A549 (tumorigenic), and H292 (tumorigenic and locally invasive)) was conducted. Candidate biomarkers for lung cancer were identified using nano flow reverse phase capillary liquid chromatography (Ncap-RP-LC) coupled to a hybrid ion trap Fourier transform ion cyclotron resonance mass spectrometer (LTQ-FTMS). These candidate biomarkers were then compared to proteins from healthy individuals and lung cancer patients using strong cation exchange (SCX) LC and Ncap-RP-LC cupled to an LTQ-FTMS. Select proteins were observed in the in vitro secretome (specific to lung cancer cell lines) as well as in human plasma from lung cancer patients.
