In quantitative bioanalysis by LC/MS/MS, one of the biggest obstacles is matrix effects on ionization. The matrix effect mechanism and its resolution have been discussed extensively in the past. There remains a challenge in finding a robust procedure for quantifying matrix effects. The traditional way of quantifying matrix effects (i.e. matrix factors) is by comparing the signal response of an analyte in the presence of a matrix with the response of the analyte in the absence of the matrix. This approach has two major drawbacks: the first is the non-specific binding of the analyte in a clean solvent; and second, the approach is not amenable to online extraction methods where extraction efficiency may mask the measurement of matrix effects. These drawbacks frequently prevent us from getting an accurate assessment of matrix effects.
