Discussed herein is a novel approach to current proteomics challenges in protein frame shift processing. Ty3 is a retrovirus-like element in Saccharomyces cerevisiae. Within the genetic makeup are open reading frames encoding homologs of retroviral Gag and Pol proteins subsequently requiring post processing prior to full assembly. Expression of Ty3 in yeast results in the formation of 50 nm particles, allowing reverse transcription to occur and Ty3 DNA to be imported into the nucleus whereby undergoing integration into host chromosomes. Identifying and understanding the mechanism of Ty3 post processing provides insight to the biochemical behavior of similar retro viruses, highlighting possible pathways for therapeutic interference.
