Viventia's product development platforms, Hybridomics~(TM), ImmunoMine., and UnLock~(TM), have been designed to identify tumor-specific antibodies and their cognate antigens. The complexity of membrane antigens hinders the possibility of tailoring purification strategies using conventional approaches. Although 2D-PAGE is capable of resolving 1500-3000 proteins in a single separation, the separation and visualization of higher molecular weight, hydrophobic membrane proteins is generally not possible with this method. Therefore, a 2D-liquid-phase separation that incorporates the properties of 2D-PAGE separation but is feasible for hydrophobic, high molecular weight transmembrane proteins would seem to be an ideal method for the separation of membrane associated tumor antigens. ProteomeLab.PF2D-MS/MS analysis from prefractionated antigen mixtures proved successful in the identification of the cognate tumor-associated antigens.
