A Simple Liquid Chromatography Mass Spectrometric Method for Quantitation of Ondansetron in Beagle Plasma using Granisetron as an Internal Standard.

Ondansetron (Figure 1) is an antiemetic drug used in the treatment of chemotherapy induced nausea and vomiting. A liquid/liquid extraction procedure (methyl t-butyl ether) was used on 0.1 ml plasma samples, with an extraction efficency of 68%. A single quadrupole electrospray mass spectrometer in positive ion mode was developed for ions 294.2 (ondanseteron), 313.2 (granisetron, Figure 2) and a unknown at 280 m/z. The assay was linear in the range of 4 to 500 ng/ml (Figure 3). Chromatograms demonstrating the LLOQ and the unknow 280 m/z are shown in Figure 4 and 5, respectively.
