摘要

The plasma proteome is of great value in disease research and therapeutics, as it contains important biomarkers and drug-related proteins. However, the complexity and wide protein concentration range of plasma make it the most difficult specimen for proteomics analysis. In large-scale proteome analysis, peptides are identified by MS and MS/MS analysis and then sorted to determine the parent proteins. As, high resolution mass spectrometry can determine an accurate molecular weight of peptide, it is useful for increasing a confidence in MS/MS based peptide identification. To get higher true positive peptide assignments from complex proteome analysis, we optimized the searching strategy by combining accurate MS with FT/MS and MS/MS analysis with LT/MS and applied to plasma proteome analysis.
