PRELIMINARY IDENTIFICATION OF SORGHUM MOSAIC VIRUS INFECTING SUGARCANE IN GUANGXI

Aims: Sugarcane mosaic disease is a popular disease. The dominating pathogens are sugarcane mosaic virus (SCMV) and sorghum mosaic virus (SrMV). In order to investigate the pathogen in Guangxi, an identification research was carried out. Methods & Results: A sample of sugarcane leaves showing chlorotic stripe and mosaic symptoms was collected from suburb in Manning. The infected sugarcane leaves were purified by one cycle of differential centrifugation and PEG double precipitation. Filamentous particles were observed under electronic microscope. ELISA test indicated that the virus isolate had positive reaction with SCMV and SrMV multiclonal antiserum and negative reaction with SCMV monoclonal antiserum. The isolate was pre-identified as SrMV. A specific primer that was designed according to the coat protein gene of SrMV was used in the RT-PCR and IC-RT-PCR to test the sample. A segment of 1.1 kb was obtained in both methods and no segment in healthy control. The segment nucleotide sequence analysisindicated that the segment contain 1010 nucleotides. Sequence homology was analyzed by using the BLAST program (http://www.ncbi.nlm.nih.gov). The result showed that the isolate had more than 95. 32% nucleotide sequence and 97% amino acid sequence whichwere homogenous with SrMV from Zhejiang and Yunnan. So the isolate was estimated as SrMV according to the classifying standard of Potyviridae. Significance of Study: This result is of guiding significance for resistance breeding to sugarcane mosaic disease and virus detection of healthy seedlings.
