Aims, to develop a biochemical and molecular characterization of resistant sugarcane somaclones by analytical isoelectric focusing of leaf protein and AFLP markers. Methods & Results: Somaclonal variation is used as an alternative resource in sugarcane breeding applications. Eye spot disease caused by Helminthosporium (Drechslern) sacchari Butler (Subram.) produce serious damage to sugarcane at specific areas. Disease resistance markers contribute to a more efficient selection procedure. The objective of this paper were the determination of biochemical and molecular differences of somaclonal variants to eye spot disease resistance. Biochemical and molecular characterization of resistant somaclones by analytical isoelectrofocusing of leaf protein andAFLP markers were used. The interaction between labeled toxin with somaclones and donor variety (C 87-51) permitted to detect specific proteins associated to the somaclonal resistance with differences in their structure and function depending on different resistance degrees to the disease. On the other hand, one of the proteins detected shown a low level of binding activity to the toxin in the resistant somaclones. Different DNA patterns between resistant somaclones and the susceptible donor variety were also detected. Additionally, AFLP markers for the identification of a genetic diversity group and somaclones and donor variety studied was also reported.
