eNOS Polymorphisms in Human Traumatic Brain Injury

Single nucleotides polymorphisms (SNP' s) of endothelial nitric oxide synthetase (eNOS) have been associated with cerebral and cardiac vascular disorders including vasospasm, infarction, and aneurysms,, We investigated the eNOS SNP's at three sites (T786-C, Glu/Asp298, VNTR (27 bp) in patients with severe traumatic brain injury (TBI).Methods: SNP analysis was performed in 32 patients with severe TBI using banked genomic material. The SNP region is amplified by PCR and then sequenced.Results: No genotype related difference in ethnicity, age, gender, severity of injury, intensity of therapy, injury type (diffuse vs focal), delayed hematoma or mortality. There was a strong trend toward increased morbidity and low tissue pO_2 with specific genotypes.Discussion: The genetic background an injured person brings to the transaction may influence propensity to complications. Knowledge of our patient's genetic background may permit individualized prognostication and tailored monitoring and intervention.
