Genetic and biochemical characterization of durum wheat Sicilian landraces.

In order to stady and to exploit the biodiversity of durum wheat ecotypes, biochemical and molecular characterization among and within 36 Sicilian landraces was carried out. The characterization of grain storage protein was carried out by polyacrylamide gel electrophoresis (SDS-PAGE) for identification of HMW and LMW glutenin subunits. Only some ecotypes showed an unvarying electrophoretic profile within the landraces; on the contrary, most of the landraces pointed out the presence of biotypes with different protein compositions. Polymorphism was found when Random amplified polymorphic DNA (RAPD) analysis was conducted on four landraces using 18 random primers.
