Assessing the genetic diversity of einkorn wheat using microsatellite markers.

A set of 62 wheat microsatellite markers, representing all seven chromosomes from the A genome, were used to estimate the extent of genetic diversity among and within accessions of einkom wheat, Triticum monococcum. From the 62 primer pairs that wereinvestigated, 33% failed to amplify einkom DNA. Of the remaining microsatellite markers, those that amplified single fragments were investigated further. Although multiplexing was problematic with discrepancies between the size of alleles in single reactions and multiplex reactions occurring with certain marker combinations, multiplex panels containing up to 6 fluorescently labelled markers were designed to survey allelic diversity. The majority of markers were polymorphic and the variation observed between accessions was comparable to that found in other studies. The level of intra-accession variation was low. Furthermore, duplicate accessions obtained from multiple genebanks were found to differ in some cases. Accessions that were homogenous from one genebank were found to contain a mixture of genotypes when analysed from another source.
